Heat input is important To address the factors of chemistry, mechanical properties and corrosion resistance, it is important to follow a suitable welding procedure that limits the heat input to the weld and stainless base material. Limiting the heat input reduces dilution of the weld deposit with the mild steel portion of the weld joint. This in turn helps maintain the alloy content of the weld deposit and its desired corrosion resistance. Other stainless steel grades may form undesirable phases that result in brittleness or poor corrosion resistance if held too long at a high temperature. Sigma phase (a brittle, intermetallic phase with high hardness) can form in some stainless grades at elevated temperatures and can seriously compromise mechanical and corrosion resistance properties. In duplex stainless steels, for example, the heat input is responsible for the balance between ferrite and austenite in the final weld and heat-affected zone (HAZ). The proper level of heat input can help to maintain the desired amount of each phase in the finished weld and base metal HAZ. Pitfalls to avoid: warping, cracking and oxidationStainless steel has a high coefficient of thermal expansion, a measure that refers to the rate at which a material expands with changes in temperature. In short, stainless steel expands and contracts more with temperature changes in comparison to carbon steel. Differences in the coefficient of thermal expansion and the thermal conductivity can cause some difficulty when welding dissimilar materials. Stainless steel will naturally want to expand and contract more as a result of the high heat seen during welding. Conversely, carbon steel (particularly mild steel) is a good conductor of heat and therefore will cool more rapidly and shrink faster as the joint cools. These differences add to the stress on the joint, created as both sides expand with heat and contract with cooling. This can cause warping or misalignment of a dissimilar metal weld. It can also cause cracking if the stresses created by differences in thermal expansion and contraction exceed the strength of either material. To address these two issues when welding stainless steel to carbon steel, avoid highly restrained joints that create high stresses as the joint is heated and cools down. If a highly restrained joint configuration is required, use modest heat input and some pre-heating to delay cooling of the joint after welding is completed. Insulating the weld joint after the last weld pass also will slow cooling and help prevent thermal stresses from cracking a joint. When it comes to protecting the material from oxidation, these dissimilar metal welds should be treated just as a stainless weld would. Open root joints should be shielded from the atmosphere on the backside of the weld (back purging). The practice of back purging, most frequently used when TIG welding, helps prevent contamination of the weld from behind the joint. Otherwise, the weld joint and stainless steel side of the weld can be damaged by oxidation, which is the result of a reaction with oxygen and nitrogen in the atmosphere. Oxidation will damage the corrosion resistance of the weld and stainless steel HAZ. To prevent this from occurring, purge the back of the joint with an inert gas such as argon, or use one of the commercially available coatings that can be applied to the back of a weld joint prior to welding. Preparation for stainless steel to carbon steel weldingProper cleaning and preparation are vital steps to help ensure successful welding of dissimilar materials. Grind the mill scale or coatings back by at least 1/2 inch on each side of the joint. Follow this task by cleaning the area with a solvent such as alcohol or acetone. These steps help get rid of grease and oil, which tend to carry the phosphorous and sulfur that are the primary causes of hot cracking. FCAW is among the most popular welding processes because of its flexibility and versatility. Metals like cast iron, stainless steel, carbon steel, high-nickel alloys, and low-alloy steels can be easily welded using the flexible FCAW process. This approach has encouraged the view that even true/false constative statements have illocutionary force (as in \"I hereby state and affirm\") and even performatives can be evaluable as true/false statements (as \"guilty\" verdict might be right or wrong). It is also often emphasised that Austin introduced the illocutionary act by means of a contrast with other aspects of \"doing\" by \"speaking.\" The illocutionary act, he says, is an act performed in saying something, as contrasted with a locutionary act, the act of saying something, and also contrasted with a perlocutionary act, an act performed by saying something. Austin, however, eventually abandoned the \"in saying\" / \"by saying\" test (1975, 123). According to the conception adopted by Bach and Harnish in 'Linguistic Communication and Speech Acts' (1979), an illocutionary act is an attempt to communicate, which they analyse as the expression of an attitude. Another conception of the illocutionary act goes back to Schiffer's book 'Meaning' (1972, 103), in which the illocutionary act is represented as just the act of meaning something. According to a widespread opinion, an adequate and useful account of \"illocutionary acts\" has been provided by John Searle (e.g., 1969, 1975, 1979). In recent years, however, it has been doubted whether Searle's account is well-founded. A wide-ranging critique is in FC Doerge 2006. Collections of articles examining Searle's account are: Burkhardt 1990 and Lepore / van Gulick 1991. If we adopt the notion of illocutionary force as an aspect of meaning, then it appears that the (intended) 'force' of certain sentences, or utterances, is not quite obvious. If someone says, \"It sure is cold in here\", there are several different illocutionary acts that might be aimed at by the utterance. The utterer might intend to describe the room, in which case the illocutionary force would be that of 'describing'. But she might also intend to criticise someone who should have kept the room warm. Or it might be meant as a request to someone to close the window. These forces may be interrelated: it may be by way of stating that the temperature is too cold that one criticises someone else. Such a performance of an illocutionary act by means of the performance of another is referred to as an indirect speech act. Speech acts are difficult to perform in a second language because learners may not know the idiomatic expressions or cultural norms in the second language or they may transfer their first language rules and conventions into the second language, assuming that such rules are universal. Because the natural tendency for language learners is to fall back on what they know to be appropriate in their first language, it is important that these learners understand exactly what they do in that first language in order to be able to recognize what is transferable to other languages. Something that works in English might not transfer in meaning when translated into the second language. For example, the following remark as uttered by a native English speaker could easily be misinterpreted by a native Chinese hearer: Although assertions, questions and orders are fairly universal, and most of the world's languages have separate syntactic constructions that distinguish them, other speech acts do not have a syntactic construction that is specific to them. A consideration of the syntactic means available for expressing the various speech acts leads us to see that even for the three basic speech acts laid out in the table above, speakers may choose means of expression other than the basic syntactic type associated with the speech act in question. Prokaryotic taxa with the potential to catalyze redox reactions in the S, Fe, and N cycles were investigated in greater detail due to their known significance in salt marsh ecosystem functioning. Putative function was inferred based on homology at the genus level with described prokaryotic species (Table S1). Prokaryotes putatively capable of nitrification (a.k.a. nitrifiers) exhibited higher relative abundance in areas colonized by the tall S. alterniflora phenotype, in comparison to areas occupied by the short and medium phenotypes (Fig. 4a). Dominant nitrifiers in the studied system included members of the bacterial genera Candidatus Nitrotoga and Nitrospira, as well as the archaeal genus Candidatus Nitrosopumilus (Fig. 4a). Additionally, a significant enrichment in taxa potentially involved in the Fe and S cycles was detected in the plant root relative to the bulk sediment (Fig. 4b, c, d). The putative Fe oxidizer of the Zetaproteobacteria, Mariprofundus sp., showed high relative abundance in the roots of the tall S. alterniflora phenotype, while Acidihalobacter of the Gammaproteobacteria was the predominant Fe oxidizer in the roots of the short phenotype (Fig. 4b). Putative autotrophic endosymbionts capable of S oxidation from the Candidatus Thiodiazotropha genus and Thiomicrospirales order preferably colonized the roots of S. alterniflora regardless of plant phenotype (Fig. 4c). Sulfur oxidizers from the Sulfurovum genus preferentially colonized the areas dominated by the short S. alterniflora phenotype in all compartments (Fig. 4c). Putative sulfate reducers of the Desulfobacterales order: Desulfatitalea, Desulfopila, and Desulfosarcina genera were enriched at closer proximities to the S. alterniflora root (Fig. 4d). Although sulfide is generally considered as a potent phytotoxin, moderately sulfidic conditions (< 1 mM) have been shown to actually stimulate S. alterniflora growth in a controlled laboratory experiment [71]. Energy conservation from sulfide oxidation in the root tissue was speculated to be the driver of increased plant primary production [68]. Furthermore, sulfide oxidation to sulfate has been demonstrated inside S. alterniflora root tissues using isotope tracers [11, 50]. However, it is still not clear what process, biological or chemical, dominates sulfide oxidation inside S. alterniflora roots. We propose that S. alterniflora shares a symbiotic relationship with S oxidizers in both the rhizosphere and root compartments. Sulfur oxidation may be mediated by not only Candidatus Thiodiazotropha bacteria, but also members of the Sulfurovum and Thioalkalispira genera or endosymbionts from the Thiomicrospirales order. Previously studied microbial species from the Sulfurovum genus and endosymbionts from the Thiomicrospirales order have been demonstrated to fix C; whereas members from Desulfovibrio, Thioalkalispira, and Candidatus Thiodiazotropha genera have been shown to perform both C and N fixation [4, 75, 89, 91]. Moreover, Crump et al. [14] studying the root microbiome of seagrass Zostera spp. found high transcript levels of N fixing and sulfur-oxidizing genes from Gammaproteobacteria species, including endosymbionts of marine invertebrates from the Sedimenticolaceae family, which includes the Candidatus Thiodiazotropha genus. Given that the S. alterniflora root zone is enriched in reduced S and its growth is limited by N uptake, we suggest that diazotrophy coupled to sulfide oxidation may be a key process that was previously overlooked. However, direct measurements of N and C fixation, and sulfur oxidation in the roots of S. alterniflora, their rates and controls, along with their relative contribution to plant growth remain unclear and require further research. Given that most studies, including this study, have inferred the coupling of S oxidation and N fixation based on gene homology and/or taxonomic placement, this interpretation should be treated with caution. We studied a gradient in S. alterniflora productivity to characterize the ecology and physiology of the S. alterniflora root-associated microbiome and its potential role in shaping plant physiological performance. In sediments from the tall S. alterniflora phenotype, higher prokaryotic biomass and more rapid microbial mineralization of organic matter were linked to greater inorganic nutrient replenishment for plant uptake. Prokaryotic communities from bulk and rhizospheric sediment associated with the tall S. alterniflora phenotype contained the highest alpha diversity, while a decline in diversity was observed in the root in comparison to the bulk and rhizosphere sediment compartments in all S. alterniflora phenotypes. A marked decrease in prokaryotic alpha diversity with high abundances and increased phylogenetic dispersion was observed in the S. alterniflora root compartment. Thus, we propose that the S. alterniflora root microbiome is dominated by highly active and competitive species taking advantage of available carbon substrates in the oxidized root zone. The high relative abundance of prokaryotic ASVs with putative S oxidation and sulfate reduction capability in the S. alterniflora rhizosphere and root suggests a rapid S cycle at close proximity to, or even inside, the root tissue. Moreover, both functional guilds were overrepresented in the S. alterniflora rhizosphere and root core microbiome. Rapid recycling of S is crucial for organic matter mineralization in anoxic marsh sediments. Thus, we propose that S. alterniflora shares a symbiotic relationship with S oxidizing bacteria in both the rhizosphere and root compartments. Sulfur oxidizers may benefit S. alterniflora plants not only by removing potentially toxic sulfide from the root zone, but also by coupling S oxidation with N and/or C fixation. The contribution to plant growth of each of these microbial processes represents a knowledge gap that warrants further research. The S. alterniflora core root microbiome was investigated. For this study, an ASV prevalence threshold was operationally defined by plotting the relative abundance and richness of the rhizosphere and root core microbiomes at 10% intervals from 0 to 100% ASV prevalence cutoffs (Fig. S8). A conservative prevalence cutoff of 60% was determined by visually inspecting a threshold in which richness remained stable at increasing cutoff values (Fig. S8). It then clusters nearby CpG sites with consistent change into regions. In the absence of SNP information, our method relies only on reads to quantify ASM. This novel ASM score compares favorably to current methods that also screen for ASM. Not only does it easily discern between imprinted and non-imprinted regions, but also females from males based on X chromosome inactivation. We also applied DAMEfinder to a colorectal cancer dataset and observed that colorectal cancer subtypes are distinguishable according to their ASM signature. We also re-discover known cases of loss of imprinting. We have designed DAMEfinder to detect regions of differential ASM (DAMEs), which is a more refined definition of differential methylation, and can therefore help in breaking down the complexity of DNA methylation and its influence in development and disease. Based on the current state of ASM detection from BS-seq reads, we set out to develop a simple yet effective method to screen for genomic regions that exhibit loss or gain of ASM between samples from distinct conditions. The methods mentioned above detect ASM in individual samples; however, they do not allow a flexible comparison between groups of samples, such as that performed in a typical differential methylation analysis [36, 37], where the goal is to find the effect of treatments or diseases on methylation, reflected as increase or decrease of methylation levels. Here, we are interested in performing such differential analysis but focusing on the effect of ASM, reflected as gain or loss of allele specificity. For this task, we introduce DAMEfinder (Differential Allele-specific MEthylation finder), an R package [38] that consists of (i) a scoring function that reflects ASM for several samples; (ii) integration with limma [39] and bumphunter [40] to detect differentially allele-specific methylated regions (DAMEs); and (iii) accurate estimation of false discovery rates (FDR). We demonstrate the ASM score and DAMEfinder on two real datasets, one based on targeted enrichment BS-seq, comparing normal colonic mucosa to cancerous colorectal lesions, and another on whole genome BS-seq (WGBS), comparing blood monocytes from healthy females and males. We have developed a scoring method that provides a measure of allele-specific methylation, and developed a method (DAMEfinder) that detects regions that display loss or gain of allele-specific methylation, by leveraging existing methods into a single framework.
